Seventh-seeded Williams, hoping to win her first Grand Slam since Wimbledon in 2003, hammered 12 aces and 26 winners.
Mirza showed plenty of spirit as she saved two match-points on her serve before Williams completed the formalities with an ace to set up a meeting with 11th seed Nadia Petrova of Russia, who beat Bulgaria's Magdalena Maleeva 6-4, 6-4.
